Замены одиночных нуклеотидов (SNP) составляют значительную долю полиморфизма ДНК практически любых организмов и оказывают заметное влияние на их жизненный статус. Детекция уже известных SNP крайне важна, что обусловливает огромное разнообразие предложенных на сегодняшний день методов SNP-типирования. Одними из наиболее ранних методов анализа SNP являются аллель-специфичная гибридизация с олигонуклеотидами, получившая развитие в виде флуоресцентно-меченных зондов, и ПЦР-ПДРФ, основанный на расщеплении ампликонов подходящими рестрикционными эндонуклеазами, на сайты которых приходятся полиморфные нуклеотиды, или конструировании праймеров таким образом, что подобные сайты генерируются непосредственно в ходе ПЦР. Кратко рассмотрены также методы детекции одиночных нуклеотидов в гетеродуплексах с помощью некоторых химических реагентов и ряда эндонуклеаз.
