The PON1 gene polymorphic marker rs662 is associated with longevity in Bashkirs

Summary:

Longevity is the complex socio-biological phenomenon characterized by human achievement the age which significantly exceeds the average population life expectancy. The possibility of achievement of longevity depends on a combination of exogenous and endogenous factors. At the molecular-genetic level, the way of their interaction is determined by the genes of oxygen and xenobiotics metabolism enzymes, which protect the body from the toxic effects of external agents. One of these genes is the paraoxonase 1 gene. Alleles of the PON1 gene polymorphic marker rs662 (192Q>R) are associated with enzyme activity and the development of polygenic diseases which limit lifespan. With the aim of the search for associations of the PON1 gene polymorphic marker rs662 with longevity in the ethnic group of Bashkirs, a comparative analysis of alleles and genotypes frequencies among different age groups was carried out; age dynamics of genotype frequencies was performed using regression analysis in general group. Among Bashkirs, PON1*Q, PON1*R alleles and PON1*Q/Q, PON1*Q/R, PON1*R/R genotypes were found with frequencies of 69.96, 30.04, 48.7, 42.51, 5.76%, respectively. The PON1 gene polymorphic marker rs662 is associated with longevity: the chance to become the long-lived person is higher among carriers of PON1*Q/R heterozygous genotype (55-100 years, OR=1,022, P=0,043).
