The miss-match reparation system maintains the integrity of the genome by correcting unpaired bases that result from the synthesis or damage of DNA, but limits the recombination between the homeological DNA sequences. Suppression of the expression MSH2 miss-match repair gene may increase the crossing over frequency between the chromosomes of different species of tomatoes. For effective RNA-interference of MSH2 genes in interspecific hybrids, knowledge of their nucleotide sequences in different species is necessary. In the present work, nucleotide sequences, partially encoding II and IV, and fully encoding the III domains of the Msh2 protein in five wild tomato species and a cultivar of the «Margolobe» variety have been established for the first time. Presence of a conserved sequence in the III domain with a key residue of arginine is shown. Comparison of nucleotide and amino acid sequences confirmed the high degree of conservatism of the MSH2 genes. The possibility of RNA-interference of MSH2 genes is discussed by using the new pro-SmAMP2 promoter, whose high activity is noted in the reproductive organs of transgenic plants.
