In vitro rooting cultivars of black currant (Ribes nigrum L.) of the Bashkir breeding

Summary:

Black currant (Ribes nigrum L.) is one of the most valuable berry bushes. The development of protocols for microclonal reproduction in vitro R. nigrum of the Bashkir selection is aimed at mass production of particularly valuable varieties and hybrids. One of the important points in the microclonal propagation is the stage of rooting plants obtained in vitro. The article presents the results of the selection of the optimal conditions for rooting R. nigrum of the Bashkir breeding in conditions in vitro. The objects of research - varieties of R. nigrum of the Bashkir breeding Karaidel and Chishma. As a control, when cultured in vitro used cultivar Seyanets Golubka. For rooting varieties R. nigrum Seyanets Golubka and Chishma favorable is a relatively high concentration of IAA 0.75 mg/l, for varieties Karaidel – relatively low concentrations of IAA 0.1-0.25 mg / l. At relatively low (0.25 mg/l) concentrations of IAA in the nutrient medium, the formation of the root system from 1.3±0.4 to 2.9±0.5 cm depending on the variety was observed in all studied varieties. The increase in the concentration of IAA from 0.75 to 1.0 mg/l in the nutrient medium led to the formation of short roots in all studied varieties. The maximal indices of escape length and number of leaves in the studied varieties of R. nigrum of the Bashkir selection were revealed in the variants of experiment with relatively high (0.5 - 1.0 mg/l) concentration of IAA in the nutrient medium. The variety Seyanets Golubki shoot growth is positively influenced by low concentrations of IAA 0.1-0.25 mg/l. For rooting of R. nigrum of the Bashkir breeding in conditions of culture in vitro it is possible to use micro-shoots length of 1.0-1.5 cm.
