Разнообразие методов молекулярного клонирования продуктов полимеразной цепной реакции

Аннотация:

Описаны различные методы клонирования продуктов полимеразной цепной реакции, включаядовольно экзотические. Проведен обзор большого числа экспериментальных работ методического плана, посвященных клонированию ампликонов в фаговых и плазмидных векторах. Все существующие методы клонирования ампликонов можно подразделить на несколько групп в виде клонирования по тупым и липким (выступающим) концам, А/Т-лигирования, рекомбинации. Причем каждый такой подход реализуется множеством способов, отличающихся использованием или неиспользованием ДНК-лигазы, направленным и ненаправленным расположением вставки, а также другими особенностями. Кратко рассмотрены преимущества и недостатки основных методов.
