Выделение, клонирование и секвенирование термостабильных ДНК-полимераз. I. Эубактерии

Аннотация:

После разработки ПЦР с использованием термостабильной Taq-полимеразы из термофильной эубактерии Thermus aquaticus интерес к таким ферментам резко возрос, и было найдено немало ДНК-полимераз из нескольких видов этого рода микроорганизмов, а также некоторых других термофильных эубактерий. Для многих из них были предложены методы выделения и очистки этих ферментов, проведено клонирование и секвенирование кодирующих их генов, сопровождающееся созданием штаммов Ecoli - продуцентов соответствующих ДНК-полимераз, в том числе их укороченных вариантов, лишенных 5’→3’-экзонуклеазной активности. При этом редактирующей 3’→5’-активности у данных ферментов за редким исключением не имеется. Наиболее широко используемым ферментом в ПЦР до сих пор остается Taq-полимераза в силу исторических причин, а также благодаря удовлетворения ею основных требований, предъявляемых при проведении классического варианта данной реакции.
