Аннотация:

В представленном обзоре отражены достижения в области генетики остеопороза, а такжесовременные подходы и направления дальнейшего исследования заболевания. Остеопороз занимает значимое место среди заболеваний с высокой смертностью и является причиной более 8,9 млн. ежегодных переломов во всем мире, характеризуется низкой минеральной плотностью костной ткани (МПКТ) и микроархитектурными нарушениями костной ткани. Распространенность остеопороза широко варьирует, достигая 5-10 кратных различий между географическими регионами, что, вероятно, обусловлено наследственным характером величины скелета и региональными особенностями рациона питания и физической активности, а также особенностями генетической структуры популяций. В исследовании генетики остеопороза началась новая эра открытий с началом полногеномных исследований ассоциаций (GWAS) сотен тысяч однонуклеотидных полиморфных вариантов (ОНП) с уровнем МПКТ и остеопоретическими переломами, идентифицированы новые локусы, ассоциированные с остеопорозом, не обнаруженные в предыдущих исследованиях кандидатных генов.
